Rare leukemia-causing protein discovered

ScienceDaily (July 2, 2012) ? Researchers at the University of Cincinnati Hoxworth Blood Center have discovered a new gene target for leukemia therapy.

These findings, slated for the July 26, 2012 print issue of Blood, the journal of the American Society of Hematology, could lead to cellular targets for a patient population that otherwise may not have desirable outcomes and could potentially stop the onset of leukemia before it begins.

A team led by Jose Cancelas, MD, PhD, an associate professor of pediatrics at the UC College of Medicine and director of the research division at Hoxworth Blood Center, found that by inhibiting in animal models the protein Vav3, which controls cell signaling, the development of this leukemia -- known as BCR-ABL lymphoid leukemia -- is delayed.

"Despite advances in the treatment of this disease, the outcome of patients with this type of leukemia is very poor because it develops resistance to standard therapies," he says. "We found that the genetic deficiency of Vav3 delays the formation of leukemia by impairing the signals from BCR-ABL and the overproduction of leukemic cells. In doing this, it also allows the standardized therapies, or BCR-ABL inhibitors, to work."

Cancelas says this finding could lead to new multi-targeted therapies where Vav3 activity is related to the formation of leukemia.

"In collaboration with Dr. Nicolas Nassar, associate professor of pediatrics at UC and a physician in the division of experimental hematology at Cincinnati Children's Hospital Medical Center, we are now trying to find chemicals with Vav3 inhibitory activity," he says. "With this knowledge, we may be able to develop a therapy that can greatly improve the lives of patients facing leukemia."

Other authors of this multinational study are Kyung Hee Chang and Susan Dunn from Hoxworth Blood Center; and Yi Zheng, John Perentesis, Abel Sanchez-Aguilera, Amitava Sengupta, Malav Madhu, Ashley Ficker, Rebecca Santho and Ashley Kuenzi from Cincinnati Children's. Collaborators also include David Williams (Boston Children's Hospital, Harvard Medical School), Michael Deininger (University of Utah), Xose Bustelo (University of Salamanca, Spain) and Xabier Agirre (University of Navarra, Spain).

This study was funded by the National Heart, Lung and Blood Institute of the National Institutes of Health, the United States Department of Defense, Alex's Lemonade Stand Foundation and Cancer Free Kids Foundation.

Continuation of this study has been recently funded by the Leukemia and Lymphoma Society of North America.

A number of the performances are plain stunning - Aran and Joan, in particular, seem born to move - and in many ways watching the movie is like watching a sporting event; there are winners and losers and favorites to cheer on.

It's an intense journey, and Kargman captures it all, down to the dancers' bloodied and bruised feet.

Yes, it is possible to create a gripping documentary about the ballet world without resorting to "Black Swan" melodramatics.

Because its subjects are so driven and so talented, "First Position," which is about ballet, is more gripping than the norm.

It's in the quick audience-reaction shots of the young dancers' exultant parents and cringing private instructors that the movie finds its most nakedly human moments.

The film shows the grueling work it takes for young dancers like Zamora to look effortless on stage, and First Position shows teachers who range from supportive to borderline abusive.

Far from the dutiful progeny of "Dance Moms," these are individuals of uncommon skill, self-knowledge, maturity and drive. They're the real deal. And so is First Position.

All of the dollars and expertise that produced this summer's blockbusters could never produce a spectacle as wondrous as 11-year-old Aran Bell dancing his heart out.

Kargman ... brings a welcome empathy to this programmatic film that follows six hopefuls as they compete in something called the Youth America Grand Prix, which we're told is the most comprehensive ballet competition in the world.

The children are almost unearthly in their agility and elegance -- until they remove their shoes, and reveal their all-too-human abrasions and injuries.

It's a wealth of material at odds with a scant running time and shallow focus.

When it's over, you won't remember so much about who won and who lost. Instead, what remains is the expression on a young dancer's face, reflected in a mirror: showing the joy of being in love with what you do.

What we experience is the overwhelming thrill of the competition and the true agony of defeat.

It could very well compete for Best Documentary at next year's Academy Awards.

For someone (like me) who has never been to a ballet, the talent of these teens and tweens is a revelation.

It's 'Spartacus' en pointe.

Inspiring documentary about hardworking young dancers.

Almost loses its sense of good taste entirely in the final act, straining to build suspense and shape heroes. The bias hangs a little ugly in an otherwise passable, somewhat enlightening effort.

The cookie cutter format Kargman uses in presenting her subjects may be unoriginal but a safe way to introduce the filmmaker to her new audience.

Child ballet dancer and former journalist Bess Kargman brings a dancer's passion and insight to her directorial debut, illuminating the addictive magic and fierce demands of a ballet dancer's life.

Really, all of the dancers' evident passion for their art is, well, spellbinding.

These kids are proud of their tights.

My name is Mary Killman, and I am half of the U.S. Olympic Duet for Synchronized Swimming. I'm a proud Native American as part of the Citizen Potawatomi Nation (Fun fact: Jim Thorpe was part Potawatomi!) I was born in Ada, Okla., but I've always called McKinney, Texas my home. Although sometimes I wonder why I claim a hometown at all -- I've lived in 42 different states so far and I'm only 21!

I've been doing synchro for 10 years, and have been on the National Team since 2007. Mariya Koroleva, my duet partner, and I are currently living and training in Indianapolis, Ind., where we are working hard and have qualified for the Olympic Games in August.

It's super exciting to be able to represent your country, and even more exciting when you realize you're one of two people representing your ENTIRE sport FOR your country! Just one month left until the Olympic rings shine brightly in everyone's minds and the Games will play constantly on millions of TVs, and I'll be one of those athletes shown! I can't wait!

I found a quote in my corrections notebook that really has hit home for me recently and I wanted to share it with everyone:

Mick Jagger walks daughter Jade down aisle

Mick Jagger made moves of a different kind on Saturday, walking his daughter, Jade, down the aisle at her wedding.

The Rolling Stones legend gave Jade, 40, away as she wed her fiance, DJ and festival promoter Adrian Fillary, according to the UK?s The Sun.

Kate Moss -- whose daughter, Lila Grace, acted as flower girl?-- was also in attendance for the happy day, the paper reports.

Goodrich Opens New Facility in California to Support Sensors and Integrated Sites Worldwide

During this event, the attendees were given a tour of design, manufacture and development areas within the new 105,000 square-foot facility. This new Brea site is a division of Goodrich Sensors and Integrated Systems business, and includes around 260 employees working in varied disciplines such as manufacturing, engineering, and management functions.

The Brea site will deliver cargo winches, rescue hoists, digital data systems and in-flight entertainment products and associated services that will greatly benefit the defense and aerospace industry. The site will function as the business' software center of excellence, through which software design and support will be offered to all global Sensors and Integrated Systems sites. The new site integrates two previously established California-based Goodrich facilities in Monterey Park and Diamond Bar. This report by Reuters indicates the role of Islam in West Africa. The growth of Islamic radicalism has increased persecution of Christians Africa. Radical Islam has also posed security threats to many countries in Africa.

06/28/2012 Nigeria (Reuters)-Al Qaeda-linked Islamists declared on Thursday they had secured full control of Mali's desert north from separatist rebels with whom they had temporarily joined forces to seize territory earlier this year [ID:nL6E8HSFJ7].

Separatist Tuareg-led MNLA rebels and armed Islamist groups, swept through northern Mali in March and April and declared an independent state of "Azawad" in the north after routing the regular troops, in disarray after a March 22 coup in the West African country.

The Tuareg separatists said they wanted an independent secular state while the Islamists said their objective was to impose sharia or Islamic law across the whole of Mali, which mostly follows a moderate form of Islam.

The relationship between the state and Islam differs significantly across western Africa, in part because of variations in the complex interaction between Islam and ethnic politics.

Here is a look at Islam in a few West African countries:

OVERVIEW:

- Islam arrived in sub-Saharan West Africa as early as the 8th century, travelling with Arab traders from North Africa. The Muslim merchants brought trade and goods to exchange for gold and facilitated trade by introducing concepts such as contract law and credit arrangements. They were valued also for the literacy they brought, which African rulers quickly discovered could help them administer their kingdoms.

- Today, countries to the north of the region such as Gambia, Guinea, Mauritania, Mali and Niger are predominantly Muslim. To the south of the region, in countries such as modern Ghana, Muslims are in the minority.

NIGERIA:

- However tension between the two communities has been a part of the landscape of Nigerian politics since independence in 1960 and problems have been exacerbated recently by the election of Goodluck Jonathan as president. He is a Christian southerner, and in the eyes of many Muslim northerners it was a northerner's turn to rule.

- The shadowy Islamist sect Boko Haram has attacked Christians, possibly hoping to trigger a religious conflict.

SENEGAL:

- A religious, economic and social force with no real parallel elsewhere, Senegal's Brotherhoods are a pillar of the moderate Sunni Islam espoused by over 90 percent of the nation who co-exist with minority Christians and others.

MAURITANIA/MALI

- In Mauritania throughout the 1990's, political liberalization allowed Islamists to participate in elections as participants and the Tawassoul party became a significant minority moderate voice. However there are more hardline Salafi activists in poorer areas.

- However Bamako has accused the Tuaregs of lending support to al Qaeda in the Islamic Maghreb (AQIM) by sharing their desert expertise and navigational skills, opening up their trade networks. It would be impossible for AQIM to operate in northern Mali without some sort of acceptance by the Tuaregs, some Sahel researchers have said.

- AQIM had its roots in Algeria's Salafist movement, but has pushed south into the lawless Sahel region in recent years where it funds operations by collecting kidnap ransoms and by siphoning off the West African drugs trade. AQIM has established an operating base for itself in the Sahara desert - spanning northern Mali, Niger, Mauritania - by exploiting the vast expanses, official corruption and weak militaries in the region.
